Y Chromosome Loss: A Possible Factor in Lung Cancer Progression and Outcomes

Insights into the impact of Y chromosome loss on lung cancer treatment outcomes may guide therapeutic choices.

Research indicates that men diagnosed with the predominant type of lung cancer are more likely to lose the Y chromosome in their cells. This phenomenon has both pros and cons; while it can prevent the immune system from combating tumors, it also enhances the effectiveness of standard anti-cancer therapies.

As men grow older, their cells frequently undergo mutations, leading to the loss of the Y chromosome. In immune cells, this loss is believed to correlate with heart disease and decreased life expectancy. Additionally, there is growing evidence that cancer cells that lose the Y chromosome may influence symptom progression, with bladder cancer being the most thoroughly researched case.

The loss of the Y chromosome is a binary occurrence—it either happens or it doesn’t. However, the health implications seem to depend significantly on the proportion of specific cells that lack the Y chromosome.

The recent study initiated by Dawn DeMeo and her team at Brigham and Women’s Hospital in Boston, Massachusetts, investigated how Y-chromosome genes are expressed in a publicly available dataset of lung adenocarcinoma samples. Lung adenocarcinoma, the most common form of lung cancer, originates from the mucus-producing cells lining the airways. Enhanced understanding of the relationship between Y loss and various health issues has motivated researchers to delve deeper into gene expression studies, according to DeMeo.

The team discovered that cancer cells, in contrast to healthy lung and immune cells, often lack the Y chromosome. This occurrence is independent of whether the tissue donor is a smoker—despite smoking being linked to lung cancer and Y chromosome loss.

The loss of Y chromosomes appears to accumulate over time. “Certain groups demonstrate a higher rate of Y chromosome loss across a greater number of cells, and we observe significant Y chromosome loss in a large fraction of tumors,” stated John Quackenbush from Harvard University.

To comprehend the reasons behind this accumulation, researchers examined other genetic alterations in Y-negative cells. They found that the loss of a common set of antigens produced by cancer cells correlates with diminished expression levels. These antigens usually notify immune T cells that cancer cells are abnormal and should be targeted. The decreased expression allows Y-negative cancer cells to proliferate unchecked.

“This implies that as tumor cells lose their Y chromosome, they become increasingly adept at evading immune surveillance, suggesting a selection of tumor cells that escape immune detection,” Quackenbush explained. T cell counts were consistently lower in samples with Y loss compared to those retaining the Y chromosome.

Positive findings emerged when researchers analyzed data from 832 lung adenocarcinoma patients treated with the immune checkpoint inhibitor pembrolizumab, a medication designed to restore the body’s immune response against tumors by reversing T-cell suppression. The analysis confirmed that Y chromosome loss was linked to improved treatment outcomes.

“Patients experiencing LOY [loss of Y] are more responsive to checkpoint inhibitors,” noted Dan Theodorescu from the University of Arizona, who found similar results in bladder cancer, establishing validation against an entirely different dataset.

However, while loss of the Y chromosome is linked to shorter life expectancies for men compared to women, existing data suggests it does not impact survival in patients with lung adenocarcinoma. Further research is needed to explore how the effects of such mutations influence survival across different cancer types, according to Theodorescu. As our understanding advances, he believes that loss of Y could eventually serve as a biomarker for clinical decision-making.

Extended Use of Melatonin Linked to Negative Health Outcomes

An analysis of five-year health data from over 130,000 adults suffering from insomnia who had been taking melatonin for at least a year indicated that these individuals were at an increased risk of being diagnosed with heart failure, needing hospitalization for the condition, or dying from any cause.

Melatonin is a hormone naturally generated by the pineal gland, aiding in the regulation of the sleep-wake cycle.

Synthetic hormones, chemically identical to natural melatonin, are frequently prescribed to address insomnia (difficulties in initiating or maintaining sleep) and jet lag.

This supplement is widely available over-the-counter in several countries, including the United States.

In the recent study, researchers identified individuals with long-term melatonin use (defined as over a year recorded in electronic health records) as part of the melatonin group.

Conversely, those without any recorded use of melatonin in their medical history were classified as the non-melatonin group.

“The perception that melatonin supplements are harmless may not be accurate,” asserts Dr. Ekenedirichukwu-Nnadi from the State University of New York Downstate/Kings County Primary Care.

“If validated, our findings could significantly impact how healthcare providers counsel patients regarding sleep aids.”

Although melatonin supplements are marketed as safe sleep aids, there is insufficient evidence regarding their long-term cardiovascular safety, prompting researchers to investigate whether melatonin use elevates the risk of heart failure, particularly in chronic insomnia patients.

Heart failure, which arises when the heart is unable to pump sufficient oxygenated blood for the body’s organs to operate effectively, affects 6.7 million adults in the United States, according to the American Heart Association’s 2025 Heart Disease and Stroke Statistics.

The researchers utilized a large international database to analyze five years of electronic health records of adults with chronic insomnia who had documented melatonin use exceeding one year.

They were compared with colleagues in the database who also suffered from insomnia but did not have melatonin recorded in their medical files.

Participants with a prior heart failure diagnosis or those prescribed other sleep medications were excluded from the study.

The analysis revealed that among insomnia sufferers, those whose records indicated long-term melatonin use (over 12 months) were roughly 90% more likely to develop heart failure over five years compared to non-users (4.6% vs. 2.7%, respectively).

Similar outcomes (an 82% higher incidence) were observed for individuals who received at least two melatonin prescriptions spaced 90 days or more apart.

In a further analysis, researchers noted that melatonin users were nearly 3.5 times more likely to be hospitalized for heart failure compared to non-users (19.0% vs. 6.6%, respectively).

Melatonin group participants were almost twice as likely to die from any cause over a five-year duration compared to those in the non-melatonin group (7.8% vs. 4.3%, respectively).

“Melatonin supplements are generally regarded as a safe, ‘natural’ solution for improving sleep quality, so the consistent and significant rise in serious health issues was unexpected,” stated Dr. Nadi.

“Since melatonin isn’t officially approved for treating insomnia in the U.S., it’s surprising that physicians would prescribe it for more than 365 days,” remarked Dr. Marie-Pierre St-Onge from Columbia University.

“While melatonin is available as an over-the-counter supplement in the U.S., individuals should be cautious about using it chronically without proper guidance.”

The research findings are expected to be presented at an upcoming conference this week at the American Heart Association 2025 Academic Session.

NASA’s Asteroid Deflection Experiment Yields Surprising and Mysterious Outcomes

Following the collision of NASA’s spacecraft with an asteroid, its orbit is expected to change gradually next month, leaving astronomers puzzled.

In 2022, the Double Asteroid Redirect Test (DART) sent a nearly 600-kilometer spacecraft towards a small asteroid known as Dimorphos.

Prior to the collision, Dimorphos completed an orbit every 11 hours and 55 minutes. Observations soon indicated that the impact shortened this orbital period by approximately 30 minutes, although in the following weeks and months, the orbital period diminished by an additional 30 seconds.

Astronomers have proposed that this phenomenon might be due to the release of small debris during the impact, which could gradually cause energy loss and result in a reduced orbital path.

Recently, Harrison Agrusa and Camille Chattanette from the University of Côte d’Azur in Nice, France, asserted that debris ejection cannot solely explain the observations.

“The boulders can be flung by Dimorphos, leading to a wider dispersion. This momentum will eventually revert and be transferred back to Dimorphos,” Agrusa explains.

Agrusa theorizes that a more plausible reason for Dimorphos’ shifting trajectory is that its impact affects its rotation in a complex manner, causing rocks on its surface to shift. The friction generated by these rocks could lead to heat production from their collisions. The resultant energy loss as heat might clarify the decrease in the asteroid’s orbital path, although this study has not modeled that aspect.

“When you rearrange materials on a surface, you’re altering the gravitational potential energy of Dimorphos itself,” Agrusa states. “This adjustment could feasibly slow down the orbital period within a month, as this would be a prolonged process.”

This highlights the challenges in predicting how asteroid orbits will alter post-impact, yet according to Agrusa, this phenomenon is less concerning when deflecting an asteroid heading towards Earth. This is because binary asteroid systems like Didymos and Dimorphos are quite uncommon, making it unlikely that similar effects will occur for a solitary asteroid orbiting the Sun.

In my view, the Stern-Gerlach experiment was a pivotal moment that introduced the findings of quantum mechanics to the scientific community. Proposed by Otto Stern and carried out by Walther Gerlach in 1922, this experiment demonstrated that atoms possess quantum properties. It revealed that electrons must adhere to quantum principles. The Stern-Gerlach experiments underscore the unusual aspects of the quantum realm, suggesting that observers seem to influence the properties that particles exhibit. Measuring the quantum characteristic known as spin appears to alter the possible spin values particles can possess subsequently. Essentially, the act of observation influences the future states of a particle.

In physics, we are conditioned to perceive ourselves as separate from the physical systems we study. This experiment challenges that separation. My experience shows that students often accept this premise as an undeniable truth. However, after contemplating it multiple times, they find it at odds with their intuitive understanding of reality. Coming to terms with these outcomes is a surreal journey—wonderful and extraordinary.

When I reflect on how to articulate witnessing the decline of American science in real-time, “surreal” is the best descriptor. It does not resemble the surrealism of the Stern-Gerlach experiment; instead, it feels like a reawakening to a tangible reality. I recognize that I once harbored an illusion about the world, but the emerging reality is intriguing and exhilarating.

Our current political climate resembles a reckoning with a previously held false sense of security. It seems that although the US government might support science today, there won’t be a transformative reality waiting for us tomorrow. Instead, the government continues to squander taxpayer-funded culture, casting it into oblivion. This metaphor carries weight: when an object crosses a black hole’s event horizon, it reaches a point of no return. The object is lost forever.

We find ourselves in a similar predicament. The universe remains to be explored, but the impairments to our research capabilities are enduring, altering the trajectory permanently. The number of master’s and doctoral programs has already diminished the availability of opportunities. Aspiring professors are not receiving the same level of training, which will affect not only future scientists but also science communicators.

The US government discards publicly funded culture, casting it into voids of neglect.

The entirety of our future appears discarded. While Donald Trump has suggested cuts to NASA’s astrophysics budget and the National Science Foundation’s physics funding, these reductions in mathematics and astronomy will not prevent global hindrances to science, and will lead to far-reaching repercussions. The US plays a crucial role as a global investor in particle physics, cosmology, and investigations into fundamental truths.

While attending the 2025 Natural Philosophy Symposium at Johns Hopkins University in Baltimore, Maryland, we were alerted to devastating budget cuts to NASA’s funding. The event started with an engaging talk by philosopher and cognitive scientist David Chalmers, followed by insights from philosopher Sandra Mitchell on human reasoning, and a discussion with theoretical physicist Nima Arkani-Hamed.

We discussed questions that have captivated humanity for millennia. What is reality, and how can one engage in researching that reality? Such events emphasize the potential outcomes when provided with the necessary time, space, and resources to explore ideas.

During this gathering, NASA’s Astrophysics department appeared to be on the brink of obliteration. The repercussions of this shift will echo for decades. In 1922, Germany stood at the forefront of scientific understanding. However, after the Nazis subverted German science for their propaganda, the field never fully recovered.

This narrative may seem like a dramatic tale about a specific group of individuals. Yet, it’s essential to remember that these discussions lead to real publications. I benefited from Government Pell Grants for economically disadvantaged undergraduates and the National Science Foundation’s Graduate Research Fellowship. The research discussed in this article has been financed through several federal agencies.

This disconcerting moment affects not only scientists in the US but resonates worldwide. Much of the science we engage with originates from American institutions, making this issue pertinent to all of us.

Google to enhance AI-generated search summaries after encountering unusual outcomes

Should you keep your job or start your own business? Should you put up with an unsatisfactory relationship, or try your hand at love another time? In each case, we have to weigh the safety of what we have against riskier but potentially more valuable alternatives.

The uncertainty inherent in these dilemmas causes many of us to become paralyzed and stagnant in our analysis, ending up staying where we are and not giving ourselves a chance to win big. In contrast, some people are easily swayed by the lure of new things. They quickly turn to gambling until they lose everything due to impulsive behavior. If any of these scenarios sound familiar, help may be on the way. Thanks to a greater understanding of our underlying cognitive biases and how to escape them, we now have evidence-based strategies to think more rationally about these challenges, so we can put our lives on the line. Starting Drug Treatment Early Improves Outcomes for Crohn’s Disease Patients

A one-year study of 386 people found that receiving advanced treatment soon after diagnosis of Crohn’s disease improves outcomes for patients.

The results of this study suggest that giving patients with Crohn’s disease intensive treatment as soon as they are diagnosed may be more effective in improving their lives, Dr. Noor said.

Parks said the extra money spent on medication would be balanced out by not having to pay for subsequent scans, colonoscopies and surgeries for people with repeated relapses.

“People with Crohn’s disease don’t want to be hospitalized or undergo surgery. They want to go out into the outside world and live their lives. Anything that speeds the path to remission. It can only be a good thing,” says Ruth Wakeman of the charity Crohn’s & Colitis UK.
