Study Suggests Common Nasal Antihistamine Sprays Could Help Prevent Community Infections

Nasal sprays available over-the-counter, historically noted for their safety and efficacy in treating seasonal allergies, could be perceived in a new light following clinical trial results released on Tuesday.

The antihistamine azelastine has been observed to have antiviral properties against various respiratory infections, including Influenza, RSV, and viruses responsible for COVID, according to a growing body of research.

Researchers from Saarland University Hospital in Germany conducted a study involving 450 adults, predominantly in their early 30s. One group of 227 participants used a nasal spray three times daily, while the other 223 received a placebo spray under the same regimen.

Throughout nearly two months, all participants underwent COVID rapid testing twice weekly. The results indicated that the incidence of symptomatic infections was 2.2% in the azelastine group, markedly lower than the 6.7% infection rate in the placebo group.

Furthermore, azelastine seemed to diminish the incidence of other symptomatic respiratory infections, as highlighted in a study published in JAMA Internal Medicine.

While researchers remain uncertain about the exact mechanism by which azelastine limits infections, they hypothesize that it may bind to the virus in the nasal mucosa.

Another possibility is that azelastine interacts with the ACE2 receptor, the primary entry point for COVID viruses into human cells, thereby preventing their attachment.

“Our findings imply that azelastine could serve as a scalable and commercially viable preventive measure against COVID, especially in high-risk scenarios such as crowded indoor events and travel,” the researchers noted.

However, the study had limitations, including that all participants were relatively young and healthy, according to the researchers.

Valz emphasized that azelastine should not replace vaccination and further research is essential before considering it as a standard precautionary measure for the public, particularly for vulnerable groups.

Dr. William Messer, associate professor at Oregon Health & Science University, found the results “rationally convincing” in terms of risk reduction, but pointed out the intensive regimen of daily sprays in the trial.

He questioned whether wearing a mask might be a simpler approach to preventing COVID infection.

“Masks can be inconvenient and bothersome, yet may be easier to adhere to than remembering to use three nasal sprays daily,” Messer remarked.

Nevertheless, he added, “I do not discourage anyone who wishes to try it.”

Other researchers are seeking more data to ascertain the effectiveness of nasal sprays in high-risk populations, such as the elderly and immunocompromised individuals who require additional preventive measures.

Dr. Peter Chin-Hong, a professor at UCSF Health specializing in Infectious Diseases, speculated that azelastine could serve as an additional COVID-blocking tool for individuals already using nasal sprays for seasonal allergies, although he believes the evidence is insufficient for broader recommendations.

“While the potential is promising, I believe now is not the appropriate time to recommend it as a COVID preventative,” he stated in an email. “For those over 65, I continue to advocate for vaccination as the primary defense against COVID.”

Nonetheless, Chin-Hong highlighted that the trial results underscore the importance of targeting the nasal mucosa in developing future vaccines against COVID and other respiratory viruses as a more effective means of infection prevention.

“Current COVID vaccines have not proven to be highly effective in preventing infection,” he remarked. “There is a need for more mucosal vaccines for respiratory viruses. While flu vaccines are widely used, ongoing efforts are being made to create mucosal vaccines for coronaviruses, necessitating continued advocacy for federal support and prioritization for these initiatives.”

Neanderthals likely developed distinct food preparation traditions that varied between groups. Excavations at two caves in what is now northern Israel indicate that these ancient inhabitants had unique methods for processing similar types of prey.

Contrary to popular belief, Homo sapiens were not the first to prepare and cook food. Evidence shows that Neanderthals, who thrived in Europe and Asia until approximately 40,000 years ago, hunted, cooked a diverse array of animals, and employed flint tools to enhance their meals with wild herbs.

For further insights into Neanderthal cuisine, see Analle Jaron from Hebrew University in Jerusalem and her team, who investigated findings at the Amdo and Kebara caves.

Located merely 70 km apart, these sites provide a unique lens through which to examine cultural differences among Neanderthals. The stone tools, remnants of food, and hearths uncovered at both locations suggest that these groups occupied the caves concurrently, possibly during winter months.

As Jaron remarks, “The same animal species were being hunted, set against a similar landscape. The weather conditions were alike, and both Neanderthal groups primarily feasted on gazelles and some fallow deer.”

However, notable differences were also present. For instance, bone evidence indicates that larger prey was favored in the Kebara cave, where more substantial animals were brought in and processed.

Jaron and her colleagues utilized microscopic analysis to study bone remnants from the sediments dated between 50,000 and 60,000 years ago, examining cut marks made by stone tools.

They discovered that while the flint tools were similar across both sites, the patterns of cut marks varied. “The cuts in Kebara displayed a greater fluctuation in width and depth, while in Amdo, they were concentrated in large clusters, often overlapping,” Jaron explains.

To determine whether these discrepancies influenced hunting practices, the researchers focused on the long bones of gazelles from both sites, which exhibited similar variations.

“We’re discussing two groups living in close proximity that both butchered the same type of meat, but it appears that at one site, the meat was processed closer to the bones.” says Ceren Kabukcu from the University of Liverpool, UK.

Past studies examining bone cut marks from later human societies indicate that the variations observed in Neanderthal butchery are not merely due to a lack of skills or difference in technique. It’s not a matter of expertise; it’s a difference in approach.

Jaron posits that these contrasts are likely a result of intentional choices regarding butchering methods. She suggests that Neanderthals in Amdo may have preferred to prepare their meat in ways that preserved it, such as drying or hanging before cooking.

“In the context of opportunistic behavior in slaughtering, one would expect the most efficient methods to be employed to maximize utility; nevertheless, it appears that cultural or social factors played a more significant role,” Jaron states. “This might relate to the group’s practices, passed down through learning and generations.”

“The existence of differences and the subtlety in technological usage in daily life isn’t entirely surprising,” notes Kabukcu. “As research deepens, we may uncover more nuances across various Paleolithic sites, even in China.”

It remains uncertain whether the caves were inhabited simultaneously or if different groups intermingled. “They could have been occupied at the same time or separated by hundreds of years. There’s no definitive way to ascertain that,” Jaron adds.

Nonetheless, she points out that the pronounced clustering of cut marks in Amdo is consistent across both the oldest and youngest sediment layers, suggesting that groups returning to the cave may have maintained similar butchering practices over centuries.

“I refuse to let it go: The man fighting to preserve the final phone box in his village”

Some users are even posting videos of people they suspect without substantial evidence, making it challenging for the community to verify these claims.

Misinformation is spreading within X and other platforms, with right-wing influencers amplifying false accusations of ballot stuffing and voter suppression. Such baseless claims are contributing to the harassment of innocent individuals, including postal workers, as seen in a viral video from Northampton County, Pennsylvania.

Experts note that this community, consisting of over 50,000 members, is employing tactics reminiscent of past online forums to propagate claims of a stolen election. These tactics were previously utilized in the aftermath of the 2020 election by groups like “Stop the Steal” on platforms such as Facebook, Telegram, and Parler.

In their attempt to bolster the narrative of a “stolen election,” these groups disseminate unverified stories to a large audience, which are then leveraged by influencers to fuel suspicions of electoral malpractice. The Election Integrity Partnership has compiled a report highlighting the dangers posed by such disinformation campaigns.

Lenny DiResta, an associate professor at Georgetown University, warns of the real-world consequences of unfounded rumors being weaponized by propaganda outlets. Ordinary individuals are inadvertently caught up in these campaigns, facing unwarranted scrutiny and harassment.

Vendetta was essentially publisher EA's best take on an engine made for a defunct WCW wrestling game that was mired in development hell and causing executives to lose their heads. At one stage, in their desperation, they even suggested that it could be salvaged and remade into an intergalactic wrestling game full of alien characters. But because Holmes and Anselmo had helmed the 2001 success NBA Street (which added a faster, cartoony, hip-hop aesthetic to basketball multiplayer and far outsold that year's bigger-budget (and much more boring) NBA Live 2002), they were deemed the perfect duo to turn what began as a vague boardroom idea (what if rappers wrestled each other?) into a fleshed-out, retail game.

This meant traveling back and forth to Japan to work with development company AKI Corporation, while also touring the US to meet with rappers signed or affiliated with legendary label Def Jam, and ultimately winning their trust. They shaped the character's mannerisms, from moves to taunts. “It's been a dream life, to be honest,” Holmes adds. “I grew up obsessed with hip-hop. As a teenager, I worked in a music store and sold a bass guitar to Flavor Flav when Public Enemy played in Vancouver. Flavor actually remembered me! It was an amazing time. Who would have thought I'd end up recording with my idols and going to Snoop Dogg's family home?”

But he acknowledges that the core concept behind Def Jam: Vendetta and its sequel, Def Jam: Fight for NY, wasn't easily embraced: “Execs had a hard time understanding the concept of a fighting game about rappers. It was a controversial concept,” says the producer, who went on to work on Halo 4 at Microsoft and 343 Industries.

“Once I explained to them that to be the best lyricist in hip-hop, it's all about arguing, dissing, and competing for the throne, they finally got it,” Anselmo reveals. “Hip-hop is full of vibrant, competitive personalities, so the character roster could have a Street Fighter II or Tekken 3 vibe.”

Def Jam: Fight for NY's concept of rappers punching each other was a huge success, and critics loved it, too, with a score of 83 on Metacritic. “We rushed to make Vendetta, but it still sold well,” says Holmes. “That success gave us the opportunity to expand on the gameplay with Fight for NY and take our time to get it right.” AKI Corporation took the ever-changing, momentum-based combat from the legendary WWF: No Mercy game, released on Nintendo 64 in 2000, and sped everything up with hyper-active Blazin' Moves (my personal favorite is Busta's backbreaker, which is more destructive than Batman's spine-crushing Bane).

The art design is somewhere between The Wire and a comic book, with an ambitious range of dynamic fighting styles (kickboxing, street fighting, martial arts, wrestling, submission). There's a storyline, with Snoop Dogg playing the cunning vampire villain The Crow, who'll hit you with a cane and cleverly taunt you with, “Do you have dental insurance?” You can wear Roc-A-Wear or Ecko tracksuits, getting increasingly better dressed as you rise through the ranks of your boss D-Mob's underground fighting syndicate, slowly earning you respect. The key mission was to choose either 128-bit's Lil' Kim or Carmen Electra as his new girlfriend, and the next day Method Man would call and ask if they'd slept together. As I recalled after recently replaying my dusty GameCube copy, Fight for NY is still exhilaratingly challenging. “This game is designed in such a way that you have to sweat and really learn something new to win!” Anselmo says with a smile. “AKI's Hiro Abe (programmer) and Hiroya Tamura (artist) were geniuses. To get good at Def Jam: Fight for NY, you had to master the gameplay and get used to losing. In modern games, fighting games are just too easy. I think that's why so many people still play our game over and over.”

And there are still a lot of people playing Fight for NY. Not a week goes by without someone playing. Holmes agrees with my theory that Def Jam: Fight for NY, with its constant depictions of MCs embroiled in diss tracks and gang politics, now feels like a documentary, especially considering how much more public conflict there has become in rap. “Rap is becoming more and more dramatic,” he agrees. “You see a lot of social media videos of rappers fighting each other, and it's interesting that the violence of the game doesn't seem so ridiculous today.”

Some will argue that a game that trivializes the conflict in hip-hop is wrong and deserves more criticism, especially with so many rappers losing their lives to violence every year. Anselmo counters: “I get that, and I don't think we could make the game right now because of drill, the environment is too tough. But Def Jam: Fight for NY was just meant to be a fantasy, escapist experience. It was like a gangsta rap version of The Warriors. It was just a little fun. Citizens in Georgia rally against proposed construction of major monkey breeding facility in their community.

Residents are expressing concerns about the facility itself and the potential for monkeys to escape, which has occurred in the past. The Gullah-Geechee community faces a threat from climate change.

The National Oceanic and Atmospheric Administration (NOAA) is providing millions of dollars to help traditionally underserved coastal communities combat climate change. This funding includes a significant stretch of 12,000 square miles of land spanning from North Carolina to Florida, known as the Gullah-Geechee Cultural Heritage Corridor. Designated by Congress in 2006, this land increases the wealth of the Gullah-Geechee by allowing communities along the corridor to utilize federal funds for programs and projects and receive technical and financial assistance from the National Park Service, all in an effort to protect and preserve their history and culture.

In April 2023, NOAA also announced over $265 million in funding for 38 new projects aimed at strengthening the climate resilience of coastal ecosystems and communities. These projects include initiatives in South Carolina to accelerate living shoreline projects in underserved communities, as well as collaborations with the Gullah-Geechee Cultural Heritage Corridor to create living shoreline projects in these communities. Additionally, the funding includes $6.2 million for programs supporting the Regional Ambassador Program for Coastlines.

NOAA-recommended projects involve hiring new staff to assist restoration organizations in building relationships with the Gullah-Geechee community, identifying resiliency priorities for community members, and establishing local relationships to support future restoration efforts. Moreover, $536,000 has been allocated to form an advisory committee.

The overlooked community of scientific detectives is now gaining attention from the research community

A group of investigators devoted to finding errors in scientific research has shocked some of the world’s most prestigious research institutions and the scientific community as a whole.

The highly publicized case of alleged image manipulation in a paper co-authored by a former Stanford University president and a leader at the Dana-Farber Cancer Institute has garnered national media attention, prompting speculation from top scientific leaders that this may only be the beginning.

“At this pace, we’re going to see another paper published every few weeks,” said Holden, the editor-in-chief of Science, one of the world’s two most influential journals. Mr. Thorpe said on site.

Investigators argue that their work is necessary to rectify the scientific record and prevent generations of researchers from pursuing futile avenues due to flawed papers. Some scientists are calling for universities and academic publishers to reform their approach to addressing flawed research.

“I understand why the investigators who discovered these issues are so furious,” said Michael, a biologist and former editor of the journal eLife, as well as a prominent advocate for reform in scientific publishing. “Authors, journals, institutions, everyone is incentivized to downplay their significance,” Eisen said.

For approximately a decade, investigators identified widespread problems with scientific images in published papers and voiced their concerns online, but received little attention. Last summer, neuroscientist and then-Stanford University President Marc Tessier-Lavigne resigned amid scrutiny over allegations of image manipulation in a study he co-authored and a report criticizing his lab culture. Since then, there has been a noticeable shift. While Tessier-Lavigne himself has not been found to have engaged in any misconduct, members of his lab appear to have manipulated images in questionable ways. In January, a blogger’s scathing post exposed questionable research by top leaders at the Dana-Farber Cancer Institute, which subsequently retracted six papers and requested corrections to dozens more.

In hisresignation statement, Tessier-Lavigne stated that the committee could not find any evidence that he was aware of any misconduct, and that he had never submitted documents that appeared to be inaccurate. In a statement from its research integrity officer, Dana-Farber stated that it had taken decisive action to correct the scientific record and that discrepancies in the images were not necessarily evidence that the authors were attempting to deceive.

“We are experiencing a moment of public awareness that really turned a corner when the Mark Tessier-Lavigne scandal unfolded, and since then the Dana-Farber scandal has been the most recent and continuous,” Thorpe said.

This long-standing issue is now receiving national attention, with the emergence of new artificial intelligence tools that are helping address problems ranging from longstanding errors and sloppy science to unethically manipulated images in photo-editing software, making it easier to spot various issues.

This increased scrutiny is prompting changes in how some publishers operate. Universities, journals, and researchers are being urged to consider new technologies, the potential backlog of undiscovered errors, and methods for enhancing transparency when problems are identified.

This comes at a challenging time in academic circles. Venture capitalist Bill Ackman, in apost last month on X, discussed the use of artificial intelligence to identify plagiarism by leaders of top universities with ideological differences, and raised questions about political motivations in plagiarism investigations. More broadly, public trust in scientists and science has steadily declined in recent years, according to thePew Research Center.

Eisen stated that he does not believe investigators’ concerns about scientific images veer into “McCarthyist” territory. “I think they’re honing in on a specific type of problem in the literature, and they’re right. That’s bad,” Eisen said.

Scientific publishing is the primary means by which scientists establish a foundation of understanding in their fields and share new discoveries with their colleagues. Before publication, scientific journals review submissions and solicit feedback from researchers outside the field to identify errors or faulty inferences, a process known as peer review. Journal editors evaluate research findings for plagiarism and conduct copy editing prior to publication. While this system is not perfect, it still relies on the good faith efforts of researchers to avoid manipulating research results.

Over the past 15 years, scientists have become increasingly concerned that some researchers are digitally altering images in papers to distort or enhance their results. The field of image integrity screening has expanded significantly since Yana Christopher, a scientific imaging expert with the European Federation of Biochemical Societies and its journals, began working in the field nearly 15 years ago. At the time, “no one was doing this, and people were in denial about research misconduct,” Christopher stated. “The prevailing belief was that it was very rare and that instances of manipulating results were few and far between.”

Scientific journals now employ entire teams dedicated to processing images and ensuring their accuracy. The number of retractions of published papers has increased significantly in recent years, with records indicating that over 10,000 papers were retracted last year, according to aNature analysis. A loose collective of scientific investigators applies external pressure, often identifying and flagging errors and potential manipulation on the online forum PubPeer. Many of these investigators receive little or no compensation or public recognition for their work.

“There’s a certain level of urgency,” Eisen stated. Ananalysis of comments on over 24,000 articles posted on PubPeer revealed that over 62% of PubPeer comments were related to image manipulation. For years, investigators relied on keen observation, pattern recognition, and a grasp of photo manipulation tools. In recent years, artificial intelligence tools capable of scanning documents for anomalies have been rapidly developed and improved.

Scientific journals are now utilizing similar technology to detect errors prior to publication. In January, Science announced that it is using an artificial intelligence tool called Proofig to scan papers undergoing editing and peer review for publication.Science editor-in-chief Thorpe stated that the family of six journals quietly incorporated the tool into their workflows approximately six months prior to the January announcement. The journal previously relied on visual inspection to identify these types of issues. During the editing process, Proofig flagged papers that had not yet been published, citing “logical explanations” for problematic images that were difficult to justify, or issues that the authors had addressed prior to publication. “Less than 1% of errors are significant enough to prevent a paper from being published,” Thorpe stated.

Chris Graff, director of research integrity at publisher Springer Nature, stated that the company is developing and testing “in-house AI image integrity software” to identify duplicate images. Graff’s research integrity department currently uses Proofig to assess papers in case concerns arise post-publication. The testing process varies among journals, but some Springer Nature publications use Adobe Photoshop tools to manually identify image manipulation and conduct experiments to visualize cellular components or general discrepancies in raw scientific experimental data.

“Although AI-based tools can facilitate and scale investigations, we still believe the human element is important in all investigations,” Graff stated, emphasizing that image recognition software is not infallible and that human expertise is necessary to guard against false positives and negatives. No tool can detect all mistakes and fraud.

“There are many facets to that process. You can never catch them all,” Thorpe remarked. “As journals, institutions, and authors, we need to do a better job of addressing this when it occurs.”

Many forensic scientists have grown frustrated that their concerns have been disregarded, or that investigations have progressed slowly with little public resolution. Sholto-David, who publicly voiced his concerns about the Dana-Farber study in a blog post, stated that the response from journal editors was so unsatisfactory that he nearly “gave up” on writing a letter to journal editors regarding the errors he had discovered. Elizabeth Bick, a microbiologist and longtime image investigator, said that if she reports image issues frequently, “nothing happens.”

While public comments on PubPeer questioning research data can stimulate discussion surrounding questionable research, authors and institutions often do not directly respond to online criticism. Although journals can issue corrections or retractions, it is generally the responsibility of research institutions or universities to investigate incidents. If the incident pertains to federally funded biomedical research, the federal Office of Research Integrity may conduct an investigation.

Thorpe stated that agencies need to assume responsibility when errors are discovered and act more swiftly to openly and candidly address what occurred to regain public trust.
