New Research Reveals How Gut Microbes Influence Human Brain Evolution

Humans have larger brains relative to body size compared to other primates, which leads to a higher glucose demand that may be supported by gut microbiota changes influencing host metabolism. In this study, we investigated this hypothesis by inoculating germ-free mice with gut bacteria from three primate species with varying brain sizes. Notably, the brain gene expression in mice receiving human and macaque gut microbes mirrored patterns found in the respective primate brains. Human gut microbes enhanced glucose production and utilization in the mouse brains, suggesting that differences in gut microbiota across species can impact brain metabolism, indicating that gut microbiota may help meet the energy needs of large primate brains.

“Our research demonstrates that microbes influence traits critical for understanding evolution, especially regarding the evolution of the human brain,” stated Katie Amato, lead author and researcher at Northwestern University.

This study builds upon prior research revealing that introducing gut microbes from larger-brained primates into mice leads to enhanced metabolic energy within the host microbiome—a fundamental requirement for supporting the development and function of energetically costly large brains.

The researchers aimed to examine how gut microbes from primates of varying brain sizes affect host brain function. In a controlled laboratory setting, they transplanted gut bacteria from two large-brained primates (humans and squirrel monkeys) and a smaller-brained primate (macaque) into germ-free mice.

Within eight weeks, mice with gut microbes from smaller-brained primates exhibited distinct brain function compared to those with microbes from larger-brained primates.

Results indicated that mice hosting larger-brained microbes demonstrated increased expression of genes linked to energy production and synaptic plasticity, vital for the brain’s learning processes. Conversely, gene expression associated with these processes was diminished in mice hosting smaller-brained primate microbes.

“Interestingly, we compared our findings from mouse brains with actual macaque and human brain data, and, to our surprise, many of the gene expression patterns were remarkably similar,” Dr. Amato remarked.

“This means we could alter the mouse brain to resemble that of the primate from which the microbial sample was derived.”

Another notable discovery was the identification of gene expression patterns associated with ADHD, schizophrenia, bipolar disorder, and autism in mice with gut microbes from smaller-brained primates.

Although previous research has suggested correlations between conditions like autism and gut microbiome composition, definitive evidence linking microbiota to these conditions has been lacking.

“Our study further supports the idea that microbes may play a role in these disorders, emphasizing that the gut microbiome influences brain function during developmental stages,” Dr. Amato explained.

“We can speculate that exposure to ‘harmful’ microorganisms could alter human brain development, possibly leading to the onset of these disorders. Essentially, if critical human microorganisms are absent in early stages, functional brain changes may occur, increasing the risk of disorder manifestations.”

These groundbreaking findings will be published in today’s Proceedings of the National Academy of Sciences.

We frequently hear about the benefits of certain foods for your microbiome and overall health. However, the exact composition of a healthy gut microbiome has not been fully understood until now. A recent study involving over 34,000 individuals has advanced our knowledge of the microbial combinations that indicate low inflammation, robust immunity, and healthy cholesterol levels.

The gut microbiome influences various aspects of health, including the immune system, aging, and mental well-being. While many home testing kits claim to analyze gut composition, their effectiveness remains questionable, as defining a “healthy” microbial balance is complex.

Earlier efforts have mainly concentrated on species diversity, under the assumption that a greater variety of bacteria is beneficial. However, since microbiomes differ significantly between individuals, pinpointing specific microbial communities linked to particular health outcomes is challenging.

“The interplay between our diet, gut microbiome composition, and health is intricate. The only way to unravel these connections is through large sample sizes,” explains Nicola Segata from the University of Trento, Italy.

To develop a comprehensive understanding, Segata and his team analyzed data from over 34,500 participants in the PREDICT program, conducted in the UK and the US by the microbiome testing company Zoe, and cross-referenced the findings with data from 25 additional cohorts in Western nations.

Among the thousands of bacterial species in the human gut, researchers focused on 661 species present in over 20% of Zoe participants. They identified 50 bacteria closely linked to health markers, such as BMI and blood glucose levels, as well as 50 associated with poor health.

The 50 “good” bacterial species (22 of which are newly identified) seem to affect four key areas: inflammation and immune function, body fat distribution, and blood sugar regulation.

Healthy participants, with no known medical issues, carried approximately 3.6 more of these beneficial species than those with health conditions, while individuals at a healthy weight had about 5.2 more species compared to those who were obese.

Researchers propose that the gut microbiome plays a crucial role in releasing chemicals that influence health outcomes, including cholesterol transport, inflammation reduction, fat metabolism, and insulin sensitivity.

Among the species analyzed, most bacteria classified as either “good” or “bad” belong to the genus Clostridium. Within this group, 40 species from the family Lachnospiraceae were highlighted; 13 showed positive impacts while 27 were linked to negative effects.

“This research identifies a subset of bacteria worth further exploring for their potential impacts on health conditions like high blood sugar and obesity,” states Ines Moura from the University of Leeds, UK.

The connection between these microorganisms and diet is analyzed through food questionnaires and data collected via the Zoe app, which suggests aiming for at least 30 different plant types each week and consuming three servings of fermented foods daily, promoting fiber intake and reducing ultra-processed food consumption.

The findings indicate that most microorganisms tend to enhance health with a balanced diet or exacerbate health issues with a poor diet. However, 65 of the 661 microorganisms exhibited an inconsistent relationship.

“These 65 bacteria highlight the complexity of our microbiome,” remarks Segata, who also consults for Zoe. “Their effects might depend on the presence of other microorganisms, specific bacterial strains, or particular dietary factors.”

This classification of “good” and “bad” bacteria enables researchers to assess an individual’s gut health on a scale from 0 to 1000, which is already being applied in Zoe’s gut health assessments.

“Think of a healthy gut microbiome as a network of chemical factories. We need a diverse range of species and a predominance of beneficial bacteria to generate health-promoting chemicals that benefit the entire body,” says Tim Spector, PhD from King’s College London and co-founder of Zoe.

Nonetheless, establishing a definitive model of a healthy gut microbiome is challenging. “Defining a healthy microbiome is not straightforward, as gut composition is impacted by diet and can shift due to environmental factors, age, and health conditions requiring long-term treatment,” adds Moura.

“We need to view our bodies and microbiomes as two intricate systems that combine to create an even more complex system,” says Segata. “A change in one element can subtly affect everything else. Understanding the causative relationships is often quite complicated.”

Segata advocates for larger studies to further clarify these links and represent a wider global population. However, once a health and microbiome baseline is set, he believes it should be feasible to recommend specific foods to optimize gut bacteria.

The Gut Microbiome: Influencing Our Personality Traits

Rats that received gut microbiomes from energetic human infants seem more inclined to explore their surroundings. This research suggests that the bacteria present in our guts during childhood may influence our personalities.

“This indicates that our microbes play an active role in emotional development, rather than merely being passive presence,” explains Harriet Scherekens from University College Cork, Ireland, who was not involved in the study.

Increasing research identifies a link between the microbial communities in our guts and various aspects of our well-being, emotions, and mood. For instance, individuals lacking certain gut bacteria types seem to face a higher risk of depression and anxiety.

While it remains uncertain whether bacteria are the cause of these emotional shifts or if the microbial communities alter in response to user actions, some evidence suggests that changes in the microbiome can influence an individual’s mood. For example, fecal transplants from depressed individuals to rats appear to induce depressive behavior. Conversely, depressed rodents receiving fecal transplants have shown improved symptoms upon preliminary examination.

To delve deeper into how the gut microbiome relates to temperament, Anna Artshinki and colleagues at the University of Turku in Finland conducted fecal transplants from infants into young rats.

Initially, the team evaluated the personalities of 27 toddlers aged 2.5 years using standard temperament assessments and an activity that encouraged play with bubble guns.

“Although we couldn’t study anxiety in 2-year-olds directly, we anticipated assessing behavioral differences, such as levels of inhibition versus sociability,” notes Artosinki.

From their evaluations, researchers classified 10 infants as energetic and 8 as inhibited and withdrawn. They then selected four energetic and four restrained infants (split evenly between genders) for fecal sample collection.

Fecal samples spiked with glycerol, alongside control samples, were transplanted into 53 22- or 23-day-old rats whose intestines had been pre-cleaned.

Artshinki’s team then subjected the rats to a variety of behavioral tests. They discovered that rats with microbiomes from energetic infants displayed a greater exploratory tendency compared to those receiving control implants or feces from inhibited infants.

To investigate how gut bacteria might influence the brain, the researchers also examined rat brain tissues for gene activity changes. This analysis indicated that rats receiving microbiomes from inhibited infants showed reduced activity in dopamine-producing neurons, a neurotransmitter linked to rewarding risk-taking behavior.

“This study effectively illustrates how the early childhood gut microbiome contributes to shaping behavioral tendencies,” Scherekens remarks. “By transferring microbiomes from children to rodents, researchers have created a valuable translation between microbes, human temperament, and brain function.”

This indicates a gut-brain pathway that impacts curiosity, reward, and motivation through the dopamine system, Scherekens adds.

Nonetheless, Artshinki cautions against overstating the implications. “Overall, adult temperament traits are strongly correlated with genetics, yet environmental factors—potentially including the microbiome—may play a role in certain behavioral distinctions.”

Artosinki emphasizes that whether microbes drive the differences in children’s behaviors remains an open question. It’s possible that children exhibiting more active traits interact with their environment and new foods in unique ways, thereby developing distinct microbiomes as a result.

Using low-calorie sweeteners instead of sugar may stimulate beneficial gut bacteria and aid in weight loss maintenance. This conclusion emerges from one of the longest studies on sweeteners, suggesting these alternatives might not be as detrimental as some previous reports indicate.

Several recent analyses have cast doubt on the health benefits of various low-calorie sweeteners. Although they are widely favored by those wishing to shed pounds, research indicates they may increase hunger, elevate blood sugar, and heighten the risk of heart-related illnesses. In 2023, the World Health Organization issued recommendations against using low-calorie sweeteners for weight management.

Nonetheless, there is a deficit of extensive research on sweeteners, particularly within the framework of a healthy diet. To investigate further, Ellen Black from Maastricht University in the Netherlands monitored their effects in individuals who substituted them for sugar. She and her team enlisted 341 overweight or obese adults in Europe and provided them with a low-calorie regimen for two months, resulting in an average weight loss of 10 kilograms.

Participants then adapted to a balanced diet with less than 10% of their caloric intake from sugar. During this weight maintenance phase, 171 participants were advised to completely forgo sweeteners, while the rest were motivated to swap sugary foods and beverages for lower-calorie sweetener options. Each participant had the option to use at least 16 different sweeteners, with no limit on their usage.

After 10 months, the group utilizing low-calorie sweeteners managed an average of 1.6 kilograms more weight loss compared to the sugar group. They also exhibited a higher presence of gut bacteria that produce short-chain fatty acids—beneficial compounds previously linked to blood sugar regulation, heart health support, and weight loss maintenance.

“This indicates that replacing sugar with non-caloric sweeteners in the diet may aid in weight maintenance,” says Braak. The outcomes of this study may vary from earlier research due to its extended duration and the examination of sweeteners alongside a healthy diet, she notes. Furthermore, prior studies often focused on just a few low-calorie sweeteners, many of which were not concurrently consumed.

Regarding the differences in gut bacteria, our grasp of the microbiome is still developing, according to Elan Elinav at the Weizmann Institute of Science in Israel. Thus, it remains challenging to decipher how the changes observed in the sweetener-consuming group will influence their health. He added that it is uncertain whether these transformations stemmed from weight loss, the intake of low-calorie sweeteners, or a combination of both.

While noticeable muscle growth, like bulging biceps, is a clear advantage of weightlifting, research indicates that strength training can also be beneficial for your gut microbiota.

No singular cluster of microorganisms ensures good health, yet certain compositions are linked with enhanced medical outcomes, including a stronger immune system and healthier aging. Key players include faecalibacterium, Akkermansia, and Rose Buria Hominis, all of which generate short-chain fatty acids such as butyric acid that support the intestinal lining and combat inflammation.

While diet is undeniably significant, studies suggest that aerobic exercise—like cycling and running—can create an environment favorable for these microbial species to thrive. This may occur via improved blood flow, hormonal shifts, or metabolic by-products associated with physical activity, including lactic acid. Research on resistance training has yielded mixed results; some studies indicate significant changes, while others show negligible effects. “Current research is somewhat limited,” notes Matieu Krauss from the Norwegian Sports Science School in Oslo.

For clarity, Sven Nahnsen, from the University of Tübingen in Germany, along with his team, studied a sizable group of 150 sedentary adults who underwent an eight-week training regimen that included a variety of resistance exercises like lateral pulls, seated rows, chest presses, and leg curls.

In contrast to past research, participants utilized an “intelligent” machine that automatically adjusted weights and range of motion based on individual performance, thereby keeping a meticulous record of progress. “The data is precise as every lift is digitally documented,” shares Nahnsen.

Prior to starting, and at intervals during the program, participants submitted stool samples, which researchers analyzed for genetic and metabolic fingerprints of commonly found gut microorganisms.

Across the group, there was no average alteration in microbiome composition post-program. The outcome seemed to vary based on each individual’s training response, showing a correlation between increased intensity and microbiota changes.

High-achieving participants who reaped significant muscle strength benefits exhibited an increased percentage of faecalibacterium and Rose Buria Hominis by the program’s conclusion. “These species were more abundant in those who gained substantially more strength,” summarized Nahnsen.

The reasons why some individuals responded better to the training compared to others remain ambiguous. One might assume it relates to their effort, yet compliance levels were similar across low and high performers. An alternative explanation could be that successful participants altered their diets following their exercise routines. This lifestyle adjustment could reasonably influence both microbiota and muscle strength. However, despite attempts to account for this through dietary assessments, researchers found no substantial differences in food intake, “a factor we couldn’t completely discount,” according to Nahnsen.

Another intriguing prospect is that the microorganisms themselves may partially facilitate the improvements in strength. In high-performing individuals, changes in the microbiome may enhance the production of short-chain fatty acids, enabling muscles to better respond to the eight-week training regimen. However, Nahnsen emphasizes that this warrants further investigation.

He aspires that this research will serve as a foundation for exploring potential mechanisms. “Our goal was to establish a ‘dictionary’ of microorganisms likely affected by resistance training to assist other researchers in uncovering the mechanisms,” he stated. Strategies to promote beneficial shifts in the microbiota post-training could represent a novel approach to enhancing exercise’s health benefits and sports performance.

It’s worth noting that the trial did not include a control group of participants who remained inactive. Nevertheless, Klaus, who did not participate in the study, is optimistic about the findings. “These results reinforce the idea that exercise positively impacts gut health, regardless of the specific type, and highlight that resistance training can induce microbiome-related changes tied to specific health benefits,” he remarked. “This underscores the importance of integrating both endurance and resistance training into health promotion strategies aimed at improving microbiome health.”

Current data indicates that 91% of individuals in the UK, and likewise in the US, do not meet the recommended fiber intake. 95% of people lack adequate fiber.

So, ramping up fiber consumption must be beneficial, right? However, it’s not that straightforward, particularly in relation to diet and health.

Intestinal Reactions

Fiber plays a crucial role in gut health for several reasons.

Though it is a carbohydrate, fiber differs from other carbs like starch and sugar; it is not digested in the small intestine. Instead, it travels to the large intestine where gut bacteria utilize it. These bacteria offer various health benefits by producing short-chain fatty acids (SCFAs) that contribute positively to our health.

Non-decomposable fibers absorb liquid, increase stool bulk, and make elimination smoother. Thus, boosting fiber intake can help prevent constipation and regulate bowel movements.

Individuals with IBS often believe enhanced fiber intake soothes symptoms, particularly if they experience more constipation than diarrhea.

The Risks of Overconsumption

The gut is a complex ecosystem, finely tuned to maintain balance, and introducing excessive specific nutrients can lead to unintended consequences. Some proponents of “fibremaxxing” advocate for fiber intakes exceeding 50g (1.8oz) daily, possibly resulting in side effects such as bloating, cramps, and diarrhea.

There’s also a variety of fiber types to contemplate. Soluble fibers absorb water and slow digestion, while insoluble fibers pass through largely intact, hastening digestion. Moreover, fermentable fibers feed bacteria in the large intestine.

A healthy gut requires a balance among these fibers. Excessive insoluble fiber (found in wheat bran and some vegetables) may bulk up stool but irritate sensitive digestive organs. Additionally, recent trends involving enemas can disrupt the natural rhythm of the intestines and eliminate beneficial bacteria.

Juice cleansing is neither a healthy nor a safe dieting approach.

Experts advise caution regarding trendy supplements and extreme elimination diets that exclude entire food groups without proper oversight, as these may reduce microbial diversity and impair digestion. Your gut won’t appreciate these drastic resets; what it truly needs is ongoing, careful, and balanced support.

Gut Microbiota Alterations Associated with Fertility Challenges in PCOS Patients

Women diagnosed with polycystic ovarian syndrome (PCOS) seem to face a heightened risk of pregnancy complications linked to lower levels of gut microorganisms.

PCOS stands as the most prevalent hormonal disorder among women of reproductive age and is a leading cause of infertility. Yet, the underlying causes and effective management remain somewhat elusive. For instance, it remains unclear why women with PCOS who do conceive are more susceptible to complications such as miscarriage, premature birth, and gestational diabetes.

Aixia Liu from Z Jiang University in China and her research team tracked the pregnancy outcomes of 220 women across 44 cities, half of whom had PCOS. All were younger than 35 and provided blood samples as well as stool and tissue samples from the endometrial (internal lining of the uterus).

Despite having comparable pregnancy rates, women with PCOS exhibited nearly double the likelihood of facing pregnancy complications, including miscarriage, premature birth, gestational diabetes, and delivering babies with low birth weight. Furthermore, researchers determined that women with PCOS have roughly half the quantity of gut bacteria, particularly Parabacteroides merdae, which was more closely linked to pregnancy outcomes than in those without the condition.

P. merdae aids in converting specific nutrients, like the amino acid isoleucine, into beneficial compounds known as short-chain fatty acids, which are crucial for reproductive health. This likely accounts for the 39% higher average isoleucine levels in the blood samples of PCOS participants and their 10% lower levels of short-chain fatty acids. Additionally, high isoleucine concentrations were noted in the endometrial tissue.

In laboratory settings, when researchers cultured the participants’ endometrial cells and introduced isoleucine, they observed increased markers of cell senescence, indicating wear or damage in the cells that compromised their function. Isoleucine seemed to disrupt the processes endometrial cells typically undergo during pregnancy.

“These findings suggest age-related changes within the uterus,” Li remarked in a statement from the annual meeting of the European Breeding Association, where the data was revealed. “Our research points to high levels of isoleucine from P. merdae potentially impairing endometrial health, even in women under 35.”

Such results suggest that alterations in the gut microbiome could play a role in PCOS-related fertility challenges; however, Andrea Dunaif at the Icahn School of Medicine at Mount Sinai, New York, expresses skepticism. Her studies indicate that reproductive aging in PCOS is more delayed than accelerated. “Reproductive issues often improve in your 30s,” Dunaif notes. “This leads me to doubt the significance of endometrial findings and their true responsibility for pregnancy outcomes.”

Nonetheless, these insights could pave the way for enhanced fertility treatments for women with PCOS. “Where might we intervene in the microbiome and adjust with various probiotics?” Dunaif suggests.
